Analysis: Dissolve the Sample in 10 mL of cold water. Add 20 mL of a cold, saturated solution of sodium chloride, add phenolphthalein, and then titrate with Titrant, keeping the temperature of the solution between 10C and 15C during the entire titration. Perform a blank determination and make any necessary correction. Each mL of Titrant is equivalent to 120.0 mg of monobasic sodium phosphate (NaH2PO4).
Acceptance criteria: 98.0% to 103.0% on the anhydrous basis

Insoluble Substances:
Sample solution: Equivalent to 10.0 g of NaH2PO4-H2O in 100 mL of hot water
Analysis: Pass through a tared filtering crucible, wash the insoluble residue with hot water, and dry at 105C for 2 h.

CHARACTERISTICS
Colourless crystals or a white powder.
Very soluble in water; very slightly soluble in ethanol (96%).
Dissolve 10.0 g in sufficient carbon dioxide-free water prepared from distilled water to produce 100 ml (solution S).

IDENTIFICATION
A. Solution S is faintly acid.
B. Solution S yields the reactions characteristic of phosphates.
C. Solution S neutralized with a 10% w/v solution of potassium hydroxide yields reaction A characteristic of sodium salts.
D. Complies with the test for Loss on drying.

TESTS
Acidity: pH of a mixture of 5 ml of solution S and 5 ml of carbon dioxide-free water , 4.2 to 4.5.
Clarity and colour of solution: Solution S is clear and colourless.
Arsenic: 0.5 g complies with the limit test for arsenic - 2 ppm.
Heavy metals: 12 ml of solution S complies with limit test A for heavy metals. Use lead standard solution (1 ppm Pb) to prepare the standard (10 ppm).
Iron: 10 ml of solution S complies with the limit test for - iron 10 ppm.
Chloride: Dilute 2.5 ml of solution S to 15 ml with water. The resulting solution complies with the limit test for chlorides - 200 ppm.
Sulphate: To 5 ml of solution S add 0.5 ml of hydrochloric acid and dilute to 15 ml with distilled water. The resulting solution complies with the limit test for sulphate - 300 ppm.
Reducing substances: To 5 ml of solution S add 0.25 ml of 0.02M potassium permanganate and 5 ml of 1M sulphuric acid and heat in a water bath for 5 minutes. The solution retains a slight red colour.
Loss on drying: When dried to constant weight at 130C, loses 11.5 to 14.5% of its weight. Use 0.5 g.

ASSAY
Procedure
Sample: Equivalent to 2.5 g of Na2HPO4
Sample solution: Transfer the Sample to a 250-mL beaker. Add 50 mL of water and 40.0 mL of 1 N hydrochloric acid and stir until dissolved.
Blank: Transfer 40.0 mL of 1 N hydrochloric acid to a 250-mL beaker. Add 50 mL of water.
Analysis: Titrate the Blank with 1 N sodium hydroxide and record the volume of 1 N sodium hydroxide consumed. Titrate the excess acid in the Sample solution potentiometrically with 1 N sodium hydroxide to the inflection point at about pH 4, and record the burette reading. Subtract this burette reading from that of the Blank and designate the volume of 1 N sodium hydroxide resulting from this subtraction as A. Continue the titration with 1 N sodium hydroxide to the inflection point at about pH 8.8, record the burette reading, and calculate the volume (B) of 1 N sodium hydroxide required in the titration between the two inflection points (pH 4 to 8.8). Where ;A is equal to or less than B, each mL of the volume A of 1 N sodium hydroxide is equivalent to 142.0 mg of Na2HPO4. Where A is greater than B, each mL of the volume (2B - A) of 1 N sodium hydroxide is equivalent to 142.0 mg of Na2HPO4.
Acceptance criteria: 98.0% to 100.5% on the dried basis.

Insoluble Substances:
Sample solution: Equivalent to 5.0 g of Na2HPO4 in 100 mL of hot water
Analysis: Filter through a tared filtering crucible, wash the insoluble residue with hot water, and dry at 105C for 2 h.

DEFINITION

Sample: 5.5 g of Tribasic Sodium Phosphate, on the anhydrous basis
Blank: 100.0 mL of 1 N hydrochloric acid, accurately measured
Titrimetric system
Mode: Residual titration
Titrant: 1 N sodium hydroxide
Endpoint detection: Potentiometric
Analysis: Transfer the Blank to a 400-mL beaker and titrate with the Titrant to the endpoint at a pH of 7.0. Record as the volume consumed and designate as A. Transfer the Sample to a 400-mL beaker, add 100.0 mL of 1 N hydrochloric acid, and stir until dissolved. Pass a stream of carbon dioxide-free air, in fine bubbles, through the solution for 30 min to expel carbon dioxide, covering the beaker loosely to prevent any loss by spraying. Wash the cover and sides of the beaker with a few mL of water.
Titrate the excess acid potentiometrically with the Titrant to the inflection point at a pH of 4. Record the burette reading and designate as B. Protect the solution from carbon dioxide absorbed from the air and continue the titration with 1 N sodium hydroxide VS to the inflection point at a pH of 8.8. Record the burette reading, and designate as C.
Calculate the amount of Titrant consumed by the Sample to the first inflection point, correcting for the Blank (V1 = A - B) and the amount of Titrant consumed by the Sample between the two inflection points (V 2 = C - B). If ;V 1 is equal to or greater than 2V 2, calculate the amount of Na3PO4 in the portion of Sample taken:
D = V 2 x N x F
V 2 = volume of Titrant consumed between the two inflection points (mL)
N = actual normality of the Titrant (mEq/mL)
F = equivalency factor, 163.9 mg/mEq
If V 1 is less than 2V 2, calculate the amount of Na3PO4 in the portion of Sample taken:
D = (V 1 - V 2) x N x F
V 1 = volume of the Titrant consumed to the first inflection point, correcting for the Blank (mL)
N = actual normality of the Titrant (mEq/mL)
F = equivalency factor, 163.9 mg/mEq
Calculate the percentage of Na3PO4 on the ignited basis in the portion of Tribasic Sodium Phosphate taken:
Result = [10/(100 - L)] x (D/W)
L = percentage calculated in the test for Loss on Ignition.
D = amount of Na3PO4 found (mg)
W = weight of the Sample (g)
Acceptance criteria: NLT 97.0% of Na3PO4 on the ignited basis. Na3PO4-12H2O (dodecahydrate) contains NLT 92.0% of Na3PO4 on the ignited basis.

Loss on Ignition:
Sample: 2 g
Analysis: Dry the Sample at 110C for 5 h, and then ignite at 800C for 30 min.
Acceptance criteria: The anhydrous form loses NMT 2.0% of its weight, the monohydrate loses 8.0% to 11.0% of its weight, and the dodecahydrate loses 45.0% to 57.0% of its weight.
Arsenic: To pass the test.
Acceptance criteria: NMT 3 ppm
Insoluble Substances:
Sample solution: Dissolve a portion equivalent to 10.0 g of anhydrous tribasic sodium phosphate in 100 mL of hot water.
Analysis: Filter the Sample solution through a tared filtering crucible. [Note: Do not use glass.] Wash the insoluble residue with hot water, and dry at 105C for 2 h.
Acceptance criteria: The weight of the residue so obtained does not exceed 20 mg (0.2%).
Packaging and Storage: Preserve in tight containers. No storage requirements specified.
